Heme synthesis in normal and leukemic leukocytes.

چکیده
Incorporation of glycine-2-14C and A-aminolevulinic acid (ALA)-4-14C into heme was studied in normal and leukemic leukocytes. Activities of ALA synthetase, ALA dehydrase, and heme synthetase in these cells were also measured. ALA synthetase activity and/or incorporation of labeled glycine into heme were demonstrated in leukocytes from patients with acute and chronic myelogenous leukemias. Normal granulocytes, normal lymphocytes, and leukocytes from a patient with chronic lymphocytic leukemia could not utilize glycine for heme synthesis. When normal and leukemic leukocytes were incubated with labeled ALA, incorporation of the ALA into heme was observed in all kinds of leukocytes irrespective of their types and stages of maturation. ALA dehydrase and heme synthetase activities were observed in these cells. It is suggested that the leukocytes would lose enzymes for heme synthesis in the process of cellular differentiation, ALA synthe tase being the first enzyme lost.

Stimulation of Sterol Synthesis in Peripheral Leukocytes of Leukemic Mice1
Bone marrow cells and peripheral blood leukocytes from leukemic AKR/J mice synthesize sterols from acetate at a vastly greater rate than the respective cells from normal AKR/J mice, i.e., approximately 20and 100-fold. The controlling enzyme of sterol synthesis, 3-hydroxy-3-melhylglutaryl coenzyme A reducÃ-ase, is readily detectable in gradient-purified leukocytes from leukemic mice, whereas nor...

متن کاملMetabolism by Leukemic and Normal Human Leukocytes in Relation to Cell Maturity and Type
The metabolism of an essential amino acid, isoleucine, by human leukemic and gradient-separated normal human leukocytes of various types and maturity was studied. Blood leukocytes were isolated and incubated with [U-’4CJ isoleucine. Separation of metabolic intermediates was accomplished by sequential extraction. The rate of isoleucine incorporation into protein by immature cells from untreated ...

(1) Synthesis of deoxythymidine by either direct transfer of deoxyribosyl to thymine (pyrimidine deoxyribosyltransferase) or by a coupled deoxynucleoside phosphorylase mechanism is approximately twofold greater with normal leukocyte extracts (55 to 88% granulocytes) than with extracts prepared from leukocytes obtained from patients with chronic myelogenous leukemia. Activities in lymphocytes (n...
